Megastrol acetate is a progestin a steroid hormone that is administered orally to treat anorexia and cachexia or serious unexplained weight loss. Also used as an antineoplastic agent to treat certain types of malignancy.Megestrol acetate is a synthetic progestogen that increase appetite and body weight in patients with cancer and AIDS and also improves quality of life in patients with cancer. MA was first synthesized in 1963, and initially developed as a contraceptive, then evaluated in the treatment of breast cancer in 1967 and later in endometrial cancer. Currently, MA is used for its orexigenic effect as well as for its benefits in terms of weight gain in cancer patients with anorexia and cachexia. In September 1993, MA was approved by the USFDA for the treatment of anorexia, cachexia, or weight loss. MA improves nutritional status by down-regulating proinflammatory cytokines as IL-1, IL-6, and TNF-alpha.

Mode of Action
Megastrol is a synthetic progestin and has the same physiologic effects as natural progesterone. Megestrol has slight glucocorticoid activity and very slight mineralocorticoid activity. The precise mechanism of megestrol’s antianorexic and anticachetic effects is unknown.
Indications
For the treatment of anorexia, cachexia, or an unexplained, significant weight loss in patients with a diagnosis of acquired immunodeficiency syndrome (AIDS). | Malnourished dialysis patients | Cancer-Related Cachexia
Dosage
160mg OD/ 80mg BD/ 40 mg 4 times a day
